Western blot was performed using CARD8 Polyclonal Antibody (Product # PA5-75635) and a 61 kDa corresponding to CARD8 full length form and 28 kDa corresponding to C-terminal fragment of CARD8 was observed across cell lines and tissues tested with testis showing higher expression when compared to other tissues. Whole cell extracts (30 µg lysate) of Hep G2 (Lane 1), K-562 (Lane 2), MCF7 (Lane 3), Jurkat (Lane 4), HEK-293 (Lane 5) in Fig. a; Mouse Spleen (Lane 1), Mouse Lung (Lane 2), Mouse Placenta (Lane 3), Mouse Testis (Lane 4), Mouse Brain (Lane 5), Mouse Skeletal Muscle (Lane 6), Mouse Heart (Lane 7) In Fig. b were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20,000 dilution) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).
